Skeletal deformities are one of the serious problems in hatchery‐reared longtooth grouper (Epinephelus bruneus). In this study, seed production of longtooth grouper was carried out in four large hatchery‐grade tanks. Osteological development of the vertebral column, caudal‐fin skeleton and dorsal‐fin supports in larvae and juveniles was described. The vertebral ontogeny started at 4.8 mm total length (TL) with the formation of neural spines 2 and 3, and the vertebral centra began forming at 5.3 mm TL. By 16 mm TL, all vertebral bony elements were formed except for the dorsal ribs. The onsets of the ontogeny of the caudal‐fin skeleton and dorsal‐fin supports occurred at approximately 5.1 mm and 3.9 mm TL, respectively, and the completion of these bony elements occurred at approximately 20 mm TL. The incidence of deformity was examined; some type of deformity was observed in 57%–68% of the specimens. These deformities mainly occurred as three types: lordosis, around vertebrae 1–5; saddleback‐like syndrome, around vertebrae 7–11; and vertebrae fusion, around vertebrae 22–24. The incidence of prehaemal lordosis was significantly higher among individuals with an uninflated swim bladder than among those with an inflated swim bladder.  相似文献

Hybrid weakness phenomena in rice reportedly have two causes: those of HWC1 and HWC2 genes and those of HWA1 and HWA2 genes. No detailed study of the latter has been reported. For this study, we first produced crosses among cultivars carrying the weakness-causing allele on the HWA1 and HWA2 loci to confirm the phenotype of the hybrid weakness and the genotypes of the cultivars on the two loci, as reported earlier. We then confirmed that these cultivars belong to Indica. Subsequent linkage analysis of HWA1 and HWA2 genes conducted using DNA markers revealed that both genes are located in the 1,637-kb region, surrounded by the same DNA markers on the long arm of chromosome 11. The possibility of allelic interaction inducing hybrid weakness is discussed.  相似文献

1. The pharmacokinetics of monensin, including half‐life, apparent volume of distribution, total body clearance, systemic bio‐availability and tissue residues were determined in broiler chickens. The drug was given by intracrop and intravenous routes in a single dose of 40 mg/kg body weight.

2. Following intravenous injection the kinetic disposition of monensin followed a two compartments open model with absorption half life of 0.59 h, volume of distribution of 4.11 I/kg and total body clearance of 28.36 ml/kg/min. The highest serum concentrations of monensin were reached 0.5 h after intracrop dosage with an absorption half‐life of 0.27 h and an elimination half life of 2.11 h. The systemic bioavailability was 65.1% after intracorp administration. Serum protein‐binding tendency of monensin calculated in vitro was 22.8%.

3. Monensin concentrations in the serum and tissues of chickens after a single intracrop dose of pure monensin (40 mg/kg body weight) were higher than those after feeding a supplemented monensin pre‐mix (120 mg/kg) for 2 weeks. Monensin residues were detected in tested body tissues, collected 2, 4, 6 and 8 h after oral administration. The highest conentration was found in the liver. In addition, monensin residues were detected only in liver, kidney and fat 24 h after the last oral dose. No monensin residues could be detected in tissues after 48 h, except in liver which cleared completely by 72 h.  相似文献

The present study was carried out to improve fruit set, yield, quality, marketability, and exportability of ‘Washington’ Navel orange. Pre-harvest foliar sprays of GA3, K, and Ca, either alone or combined, were applied at full bloom. The following treatments were applied: {control (T1), 25 mg/L GA3 (T2), 2% K2SO4 (T3), 25 mg/L GA3 + 2% K2SO4 (T4), 25 mg/L GA3 + 2% Ca Cl2 (T5), 2% K2SO4 + 2% Ca Cl2 (T6), and 25 mg/L GA3 + 2% K2SO4 + 2% Ca Cl2 (T7). All treatments improved fruit set, yield, physical and chemical characteristics, mineral content of leaves and fruit rind, and the percentage of exportable fruit in comparison to the control. Fruit set, fruit detachment force, peel firmness, and the mineral content of leaves and fruit rind were enhanced significantly with the combination of GA3 + K2SO4 + Ca Cl2 (T7) in comparison to all other treatments and the control. Results indicated that all foliar combination treatments improved fruit quality and marketability, and increased the percentage of exportable fruit and reduced the defects of non-exportable fruit.  相似文献

ABSTRACT:   The genetic diversity of wild and hatchery-released Pacific herring Clupea pallasii collected from three brackish lakes and two bays in Honshu and Hokkaido, Japan was examined with five microsatellite loci. All loci showed high genetic variability with expected heterozygosities ranging from 0.815 to 0.945. Significant differences in genotypic and allelic distributions were detected among all locations except for between the two bays in Honshu Island. Pairwise population analysis based on the F ST values showed close genetic relationships among the locations in Hokkaido Island, and the hierarchical analyses of molecular variance showed significant genetic difference between the two islands. Those results suggest the existence of subpopulations due to natal homing. In addition, stocked fish showed as much genetic diversity as the wild fish. The pairwise population analyses also showed close relationships between the hatchery fish and the wild fish in respective stocking areas, showing that no effects of stocking programs on genetic diversity of wild populations were detected.  相似文献

1. The pharmacokinetics of difloxacin were investigated in healthy and E. coli-infected broiler chickens following intravenous and oral administration of a single dose of 10 mg/kg bodyweight.

2. After intravenous injection of difloxacin, the serum concentration–time curves were best described by a two-compartment open model. The distribution and elimination half-lives (t0.5α) and (t0.5el), respectively, were 0.10 ± 0.016 h and 3.7 ± 0.08 h in healthy chickens compared with 0.05 ± 0.005 h and 6.42 ± 0.71 h in E. coli-infected birds. The volumes of distribution Vdss were 3.14 ± 0.11 and 9.25 ± 0.43 l/kg, with total body clearance (Cltot) of 0.65 ± 0.018 and 1.14 ± 0.1 ml/kg/h, respectively.

3. Following oral administration, difloxacin was absorbed with t0.5(ab) of 0.57 ± 0.06 and 0.77 ± 0.04 h and was eliminated with t0.5(el) of 4.7 ± 0.34 and 3.42 ± 0.19, respectively, in normal and infected chickens. The peak serum concentrations were 1.34 ± 0.09 and 1.05 ± 0.06 µg/ml and attained a Tmax of 2.27 ± 0.07 and 2.43 ± 0.06 h, respectively. The systemic bioavailability of difloxacin following oral administration was 86.2% in healthy chickens and 90.6% in E. coli-infected birds. The minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC) of difloxacin against the field strain of E. coli O78 in vitro were 0.02 µg and 0.04 µg/ml, respectively.

4. These results show that administration of a therapeutic dose of difloxacin is effective in the treatment of E. coli infection in chickens. The serum concentration of the drug was much higher than the MIC of the E. coli O78 strain in both healthy and infected chickens.  相似文献

Summary The release of extremely early maturing varieties has made it possible to cultivate rice in Hokkaido (NL45-42°) in Japan, the northern limit region of paddy rice cultivation. Until then, rice cultivation in this region has been impracticable due to the climate condition, especially short summer and long-day more than 15 hours during summer. Experiment results confirmed that the success of rice cultivation in this area depends on raising photoperiod insensitivity varieties with short basic vegetative growth period. Moreover, in this study, the genetic factors controlling the photoperiod insensitivity of Hokkaido varieties were analyzed by using 8 kinds of tester lines for three loci, E1, E2, and E3, controlling photoperiod sensitivity. It was found out that all the varieties examined carry el, a photoperiod insensitivity allele of E1 locus, but as for the other loci, E2 and E3, the existence of plural alleles were recognized. We have already clarified that almost all the japonica-type varieties grown in Japan (except Hokkaido) and Taiwan carry E1 bringing about strong photoperiod sensitivity. Accordingly, it can be concluded that el is the gene essential to Hokkaido varieties: the interchange of E1 with e1 enabled rice cultivation under long-day condition.  相似文献

The localization of metallothionein ( MT ) in the seeds and roots of soybean was investigated by immunohistochemistry. The germinating seeds at 2 hr, 1, 2, 3, 4 and 6 d including 1-mo root tips of soybean ( c.v. Toyosuzu ) with and without heavy metals ( Cu 400 μg Lor Zn 3 μg ml?1) treatment were used to demonstrate the localization of MT by the indirect immunoperoxidase technique using polyclonal rabbit antirat MT conjugated to ascaris as a primary antibody. Metallothionein was localized in the proliferating regions such as the embryo in seeds, and root and shoot apices of both the control and heavy metals-treated plants. The intensity of MT staining in the proliferating regions generally increased as the soybean seeds germinate. Starting at about 1 day after germination, MT was found in the veins and vascular bundles suggesting its translocation to other organs. Similar observation hold true in the case of plants treated with heavy metals. This means that heavy metals treatment had no effect on MT localization. However, the heavy metals-treated plants showed higher concentration of MT over the control with respect to the growth stage of soybean seeds. These indicate that MT found in soybean plays a physiological role in heavy metal transport, detoxification and cell division in a similar manner to mammalian MT.

Qualitative and quantitative Polymerase Chain Reaction (PCR) systems aimed at the specific detection and quantification of common wheat DNA are described. Many countries have issued regulations to label foods that include genetically modified organisms (GMOs). PCR technology is widely recognized as a reliable and useful technique for the qualitative and quantitative detection of GMOs. Detection methods are needed to amplify a target GM gene, and the amplified results should be compared with those of the corresponding taxon-specific reference gene to obtain reliable results. This paper describes the development of a specific DNA sequence in the waxy-D1 gene for common wheat (Triticum aestivum L.) and the design of a specific primer pair and TaqMan probe on the waxy-D1 gene for PCR analysis. The primers amplified a product (Wx012) of 102 bp. It is indicated that the Wx012 DNA sequence is specific to common wheat, showing homogeneity in qualitative PCR results and very similar quantification accuracy along 19 distantly related common wheat varieties. In Southern blot and real-time PCR analyses, this sequence showed either a single or a low number of copy genes. In addition, by qualitative and quantitative PCR using wx012 primers and a wx012-T probe, the limits of detection of the common wheat genome were found to be about 15 copies, and the reproducibility was reliable. In consequence, the PCR system using wx012 primers and wx012-T probe is considered to be suitable for use as a common wheat-specific taxon-specific reference gene in DNA analyses, including GMO tests.  相似文献
